The adenylate cyclase catalyses the conversion of ATP to cyclic adenosine monophosphate (cAMP). cAMP acts as an intracellular signalling molecule and, through interaction with the cAMP receptor protein (CRP), participates in the global regulation of gene expression in response to nutrient and environmental conditions. The cAMP-CRP regulatory system controls numerous cellular functions, including carbohydrate uptake and metabolism and the expression of proteins involved in bacterial physiology and adaptation.

The cyaA gene (also referred to as cya in earlier literature) is one of the two genes modified to attenuate the Salmonella enterica serovar Typhimurium strain used in the live vaccine AviPro® Megan® Vac 1; the other modified gene is crp, which encodes the cAMP receptor protein.
Disruption of cyaA, together with disruption of crp, interferes with the cAMP-CRP regulatory system and results in attenuation of S. Typhimurium. Studies of Δcya Δcrp strains have shown reduced virulence while retaining the capacity to colonize host tissues sufficiently to induce a protective immune response, providing the basis for their use as live attenuated vaccine strains.
Whole-genome sequence analysis of AviPro® Megan® Vac 1 confirmed disruption of the cyaA gene, corresponding to locus STM3939 in the S. Typhimurium LT2 reference genome.
